Stock Mikuni carb? What year is the bike? What mods, pipe, porting etc. have been done? and we need to Know what pilot and main jet you have in it now. The run away with the choke off sounds like you may have an air leak so it runs away, and on throttle it's not getting enough fuel because it's really lean cause its sucking unregulated air. Check the clamps on reed cage side of the carb and make sure its tight, also make sure the reed cage bolts are tight. If you can try to do a leak down test, it will tell you if you have an air leak and were. Also check for cracks or holes in the boot.
If brains were gasoline, you couldn't power a **** ant's go-cart 2 laps around a cheerio.
